I'm spending one night in San Ignacio. The next day, I would like to see the caves at Actun Tunichil Muknal and make it to Ambergris Caye by bus. Is this doable?

I do not know how long it takes to get to that cave sight, but it takes acouple of hours to get to Belize City... much longer on the local bus. First Class bus is definately worth the extra couple of dollars. Last flights to San Pedro from Belize Municipal are around 6PM. Last water taxi's I believe around 5PM. Do not run late and get stuck in Belize City unless you can stay at the Princess Hotel or maybe the St George Radisson. Belize City does have "street crime" at night.

Nope, not do-able that way. You'll have to arrange for a transfer that will drop you at the Municipal airport for the last 5:30 flight back to Ambergris. Or take the bus and spend the night in Belize City and travel to Ambergris by water taxi or plane the following day.

Sorry. I just realized what I said. The bus to Ambergris thing. Not an easy drive. Is it possible to arrange late tranportation to the caye? It looks like 5-5:30 is the end of scheduled service. Whether it's boat or plane.
